Overview

Chemokine Therapeutics Corp. (TSX: CTI OTCBB: CHKT) is a biotechnology company developing drugs in the field of chemokines. Established in 1998, we have become a leader in the design and development of peptide based drugs targeting validated chemokine receptors.

 

Chemokines are a family of small, soluble proteins that play a range of important functions in the human body, mainly in relation to the immune system, including white blood cell mobilization and in the initiation of immune responses. Among other functions, chemokines are responsible for blood cell formation through stem cell growth and differentiation.

In addition to their natural functions, chemokines play an important role in a variety of prominent diseases, including:

  • cancer, both at the level of blood vessel generation and the spreading of cancer known as metastasis;
  • viral infections such as HIV; and,
  • autoimmune diseases.

By inhibiting the action of chemokines through the use of chemokine antagonists it may be possible to reduce the severity of those diseases. In contrast, some chemokines play important roles in the body to maintain normal functioning of various organs and cellular processes. A chemokine agonist is a compound that mimics the activity of natural chemokines.

 

Drug Candidates

Our first lead drug candidate, CTCE-9908, is an antagonist of the chemokine receptor to stromal cell derived factor-1 known as SDF-1. SDF-1 is the only known naturally occurring chemokine which binds to the CXCR4 receptor present on cancer cells. This binding process is believed to be the process that causes cancer cells to metastasize (or spread) to other locations in the body from the primary cancer tumour. Approximately 90% of cancer deaths are due to metastasis. We believe CTCE-9908 interferes with the metastatic process of many types of cancers. We are currently conducting a phase Ib/II clinical trial to assess safety and preliminary efficacy in cancer patients. The phase Ib/II preliminary results released in March showed the drug to be safe and well tolerated at all the administered dose levels and we saw early evidence of a response to CTCE-9908. The final results are expected in April 2008. We have received approval from the FDA to conduct a Phase II study in the US which is planned to begin in Q3/2008.

 

Our second lead drug candidate, CTCE-0214, is an agonist of the chemokine receptor to SDF-1, present on blood stem cells. We believe CTCE-0214 will contribute to a rapid increase in the number of white blood cells (WBC) for patients with chemotherapy induced neutropenia (low WBC count). WBC's are important for fighting infection. CTCE-0214 is targeted at the multi-billion blood disorder market. We have completed a phase Ia clinical trial assessing safety in healthy volunteers and have a second phase Ib trial underway. We have completed single and multi-dose phase I studies assessing safety and preliminary efficacy which has shown a 300% increase in white blood cell counts.

Other drug candidates are in the research phase and are being developed for vascular disease, wound healing, and stroke.

Drug Discovery Approach

The majority of natural chemokines are not suitable for use as therapeutic drugs due to their instability, and potential side effects such as allergic reactions, fever and bone pain. However our team of chemists and biologists has developed an approach to discover drug candidates by generating small versions (analogs) of natural chemokines that copy their function or inhibit their function without the potential side effects and instability of natural chemokines.  

 

We synthetically produce chemokine analogs believed to have important therapeutic properties that can address potentially large markets. We first identify novel chemokines and determine their involvement in human diseases and then rapidly and economically generate analogs of chemokines that can mimic the function of their natural counterpart or inhibit their harmful effects. Using a technology known as solid phase synthesis as well as organic chemistry, we design and build synthetic peptides, forming new potential drug candidates.
